Identify particles from a particular view

Hi everyone,

I have a question in regard to viewing particles from particular orientations in cryoSPARC.

From the viewing direction distribution graph (please see attached), I learnt that the majority of my particles adopted views of 0 and 180 °, with only a few images representing views from 90 °. But are there ways in cryoSPARC that allow me to see what the particles at 90 ° look like? Knowing so would probably help me pick more particles from those particular orientations.

Thanks in advance.

Zhen

Hi Zhen,

I think that would be great! I suggested something more or less similar a while back: Orientation distribution plot suggestion [Feature request]

In the meantime I think the easiest way is to use the “Create templates” job (assuming you have a 3D model) to generate evenly spaced projections, and compare these to your 2D classes.

Cheers
Oli

Hi Oli,

Thanks a lot for your reply.

Unfortunately, there isn’t much information about the 3D structure of this protein.

Hope the cryoSPARC can implement this feature in the future

Zhen
