Questions about heterogeneity after particles expansion

Hi,

I’m working with a structure with D2 symmetry. After symmetry expansion and local refinement, I got a 4A overfitting map (below is part of the map, the helix of RNA is visiable but the details are missing).

I tried 3D classification, but it always result in evenly distribute classes.

And local refinement of these classes do not have any improvment. Is there any other ways to solve the heterogeneity after particles expansion?

Looking forward to your advise,

Thanks!

3D class needs several special parameter changes to sort well. Search bb for them, this should work the best for this case.

ab initio multi class

Het refine with several classes provided

3dva

Thanks for your suggestion, I’ve tried a few parameter combination for 3d class, include Filter resolution, Initialization mode, initial low pass, Number of classes, force hard classification.

But it seems not work really well. Also, I use symmetry expanded particles, so abinitio and het refinement seems not appropriate? I will try 3DVA with it.
