I have a problem in alignment of my particles.

my membrane protein is about 300 kDa encapsulating inside a liposome

any input, I highly appreciate your help! thanks

More information needed - what is the problem, what does your data look like, where are you running into trouble? Welcome to the forums!

have just start using CryoSparc. Data collected in Kryos and tried to 2dClass averaging. But it seems that I am not figuring out how to align the particles correctly. Thanks for your help

A blank CryoSPARC interface and unresponsive job builder

this is a challenging target which will make it difficult to understand the principles of using the program to solve structures. I would start with tutorial datasets or straightforward datasets of others on your system.

Hi @mhmd2, and welcome to cryo-EM and the forum!

I know it’s exciting to jump right in with your own data, but we strongly recommend starting with our introductory tutorial, which should only take a few hours to complete. The reason for this is twofold:

  1. The step-by-step instructions will help you get more comfortable navigating CryoSPARC
  2. If you run into any problems with your own data, it might be a problem with the data itself or how you’re processing it. If you’re working on one of our tutorials, you know that the data is good and can focus on the processing.

Once you’ve worked through that case study, you might want to check out some of our other educational material:

If you prefer videos, I’d recommend starting with this playlist, which is a recording of a workshop we ran at S2C2 in 2024. It covers a lot of the theory and practice of cryo-EM and serves as a great jumping off point!

Please feel free to always come back and ask any questions you might have!
