New case study: Pseudosymmetry in TRPV5 and Calmodulin (EMPIAR-10256)

Hello CryoSPARC users!

Today we added a new case study to the CryoSPARC guide. This case study uses images of calmodulin-bound TRPV5 to explore different ways of handling pseudosymmetry in single-particle analysis.

The case study includes practical considerations for using symmetry relaxation and 3D Classification when processing a pseudosymmetric particle. Additionally, we discuss why symmetrical (or nearly symmetrical) particles present unique challenges in single particle analysis. All users, even those who solely study asymmetric targets, may still benefit from the investigation of how changing various parameters of 3D Classification affects the results.

This case study presents the same data as the TRPV5 S2C2 workshop section, but includes more detail in some areas which were cut from the workshop for time.
