New case study: Discrete and Continuous Heterogeneity in FaNaC1

Hello CryoSPARC Users! Today we added a new case study to the CryoSPARC guide.

This case study uses EMPIAR 11631 and 11632 (FaNaC1 in the apo and FMRFa-bound states, respectively), to investigate techniques for resolving discrete and continuous heterogeneity. Topics covered include 3D Classification, Heterogeneous Refinement, 3D Variability Analysis, and 3D Flexible Refinement. At each stage, we explain the theoretical background underpinning the job type as well as practical considerations when setting up these job types.

This case study presents the same data and follows a similar exploratory path to the two FaNaC1 sections of the 2024 S2C2 workshop, but includes more detail in some areas which were cut from the workshop for time.

This is another very important topic, and I am excited to learn more about it. When faced with continuous heterogeneity, I have mostly felt like I was improvising, so I am very interested to read about a principled way to approach this problem.
