Inquiry regarding heterogenous refinement

Hi all,
I was solving a protein-DNA complex structure. I created mask for each particular segment using NU refinement volume as template and ran local refinement. I was running the heterogenous refinement job using the volume created from local refinement job and the particle stack from the previous NU refinement job. However my resolution is not great (though it is showing 4.93 A). Since I am new to Cryosparc any suggestions would greatly help me.

Hi Samrat,

It is very hard to guess what could help improving the map(s) based only on the information you are giving. If you have a good number of particles (on the ~500 k or more), you can try to understand the flexibility and variability of the sample by running 3DVA and 3DFlex. Depending on the results, it might help better classifying the particles and defining masks for local refinements.
