GSFSC does not return to basic line

Hi,

Has this ever happened to anyone? As showed below, the GSFSC curve does not return to basic line.

Are there any parameters that can be optimized.

Hi,

It is likely that you have duplicate particles, or have run a refinement with global searches on a particle set after symmetry expansion. What happens if you run Remove Duplicates?

Thank you answering. I run ‘Remove Duplicates’ and everything looks right. Now the GSFSC curve is quite normal (See attached).

Dear Oli,
I have similar problem as posted by Huang.
I run a consensus NUF refine with C1. Then performed symmetry expansion wtih C2 followed by 3DVA with mask from consensus refine in cluster mode. The separated clusters are then input into heterogeneous refine. I run into problem with the FSC curve after I try to do NUF (C1) with the particles from heterogeneous refine.
I therefore wonder:

  1. if I run a remove duplicate job, can I then perform global search refine?
  2. to generate the consensus map for 3DVA, do I need to run with C2 enforced?
  3. after particle expansion, should I run again consensus refine with expanded particles before proceeding to 3DVA?
