Hello CryoSPARC users!
Today we’ve added a new case study to the CryoSPARC Guide, focused on processing a dataset containing acetogenin-bound mitochondrial complex I from EMPIAR-10927.
We think this case study will be particularly useful for users working with sample heterogeneity, as this dataset contains discrete heterogeneity that can be revealed depending on the data processing strategy employed. Different scientific questions require different approaches, and this dataset provides an excellent opportunity to explore how alternative workflows can uncover distinct structural insights.
The case study walks through several strategies for assessing particle stack composition, interpreting job outputs, and making processing decisions to identify and characterize heterogeneous populations. Many of the key concepts and workflows are also summarized in our latest CryoSPARC Blog post, which provides a broader overview of practical approaches for handling heterogeneous cryo-EM datasets.
