Hi all, Each of my protein units is adhered to a tube based on specific helical parameters. I want to know if cryoSPARC can implement symmetric extension for each protein unit according to helical parameters, and later I would like to do local refinement on the protein units rather than the entire helical tube.

Yes this is possible, and is documented in the symmetry expansion documentation:
And yes - after performing symmetry expansion using your refined helical parameters you can then perform local refinement (or 3D classification with a focused mask, if you need to clean up your dataset further)
