Related to averaged out the DNA density during 2d classification

Hi,
I am trying to solve the structure of DNA binding protein.I am able to see the DNA intensity on micrograph.But during 2d classification,I can’t see density of DNA.Could you please suggest what can I try to get the density corresponding to DNA in 2D classification.

Hi Priyanka,

Can you post a few representative 2D classes and the parameters that you are using in the 2D classification job? That may help figuring out what’s going on.

I am using default parameter except changing particle diameter.

Hi Priyanka,

As Steve says, it is going to be difficult to help without seeing pictures of the troublesome 2D classes (and maybe a micrograph where you can indicate the DNA that you expect ought to appear in them).
