I’m trying to process negative stain data. After I imported my mrc files ( toogled as negative data), I did a blob picker job. But all target particles (white particles) were not picked. Do I have to invert my mrc files’ contrast before import into cryosparc? Is there a more convenient way?
Hi Wendy,
I had the same problem when processing negative data where there was a strong black halo around the particle and where the stain was too thin. It seemed to pick correctly on grids where the background was significantly darker than the overall particle.
