Motion correction with/without variable dose

Dear All,

I wanted to ask if there is any general consensus on whether one should allow or not allow variable dose when doing motion correction,

Does it depend in the microscope/camera setup?
Should I just try both and see which one yields a better result?

All the best,
Giovanni

It’s for if you use a variable dose rate during collection, e.g. 0.5 e-/Å^2/frame for 20 e- and then 2e-/Å^2/frame for additional 40 e-. The idea is to finely fractionate while radiation damage is low, and coarsely fractionate after high resolution information is likely lost, in order to accelerate collecting to a certain total dose. If you haven’t configured this kind of data collection, you don’t need the option.
