I’m having trouble with patch motion correction for a dataset obtained from the Krios super-resolution mode. There’s no error during patch motion correction, but when I pick particles and run 2D classification, the classes are blurry and there is some background noise.
However, in RELION, the problem doesn’t seem to occur. Any ideas on how to solve this on cryosparc?
Example micrograph output from both CryoSPARC and RELION would be helpful. That motion plot, to me at least, looks like most of the micrograph was very thin ice which melted during acquisition under exposure to the beam. Maybe the top left is thicker ice, or carbon (more likely thicker ice).
But I’d be interested in seeing what the same micrograph looks like between the two programs if the results are as different as you say!
