Local orientation diagnostics?

Hi,

Orientation Diagnostics is super useful and very fast.

Would it be possible to implement a subset of these analyses for local anisotropy analysis, similar in concept to MonoDir (Measuring local-directional resolution and local anisotropy in cryo-EM maps | Nature Communications)?

I would imagine an approach similar to local resolution estimation might be possible, using a spherical mask to calculate a masked set of cFSCs at each point within the overall molecular envelope, then reporting the relative signal or cFAR at each point. This would be potentially useful for quantifying local anisotropy (often seen in cases of interdomain hinge motion).

Cheers

Hey @olibclarke

Apologies for the delayed response – this is very interesting. Have you ever done something like this manually (via the mask input & several jobs)?

Hi Valentin,

Yes I have - I would say that qualitatively at least it matches up with my expectations based on inspection of the map

Cheers
Oli
