It’s been a while since I’ve tried the local motion correction, but I recently was re-processing one of our old ApoFe datasets with the update, and saw local motion correction doing some weird stuff. The data was K3 super-res data f-cropped by 1/2 and the gain was y-flipped, if that makes any difference. It looks like a coordinate system problem to me, but that’s just a guess. Below are 2D classifications of particles before/after local motion correction:
should have read the docs more closely, the box size to use is pre-resampled, and then is separately resampled after as needed… I’ll give that a try and update here if that was the problem: just me being an idiot!
