How to match CTF parameters from CryoSPARC-corrected micrographs to particles imported from a RELION extraction

To summarize my workflow:

I performed motion correction, CTF estimation, and micrograph curation in CryoSPARC. Then, I manually picked the start and end coordinates of my filaments using EMAN2, and saved those coordinates.

Before returning to CryoSPARC, I imported the filament coordinates into RELION and performed particle extraction there. This was done so I could generate the appropriate .star file needed to import the particles back into CryoSPARC.

The issue arises when I import these particles into CryoSPARC and attempt to link them with the curated micrographs. The CTF values are not being recognized or carried over.

Does anyone know how to resolve this issue or ensure the CTF information is properly applied to the imported particles?

Thanks in advance,
Sergio Barata

Hi Sergio, have you tried this:

Hi Olibclarke,

Thanks, Yes I tried reassing particles to micrographs and right afterwards run the refinement (It did not work). I see that the tutorial recommend you to rerun a Particle extraction via Extract From Micrographs, is this mandatory for the further jobs to work and recognize the CTF values?

Thanks!

Yes, you will need to either re-extract from the micrographs, or provide the path to the particle stacks when you import the particle stack from the star file.

Thank you!! It worked now
