Highpass filter for micrograph visualization?

Hi,

Would it be possible at some point to add an interactive highpass filter (maybe a fixed frequency, e.g. 150Å) to Manual Picker, to complement the existing lowpass filter slider?

In cases of “bumpy” or uneven ice, applying a ~100-200Å highpass filter can be helpful to flatten out the uneven background, making manual picking easier. This can be particularly the case after denoising, where otherwise imperceptible features of the background can become more prominent.

Thanks Michael! It would also be great to have a highpass (lower res limit) option in 2D classification - currently one can do this by using the downsample particles tool to generate a highpass filtered set, but it would be useful to have it built in

Thanks for the request! Highpass filtering in 2D classification should be available in a (not too distant) future version of CryoSPARC.

Thanks for the bump. Just to confirm, are you referring to highpass filtering in Manual Picker? (The use of a Minimum alignment resolution (Å) was added to 2D Classification in CryoSPARC v4.4.)

by the way, the power / NCC score can only be shown in the inspect particles after blob pick. when I do manually pick after blob pick then inspect particles, there is no power / NCC score selection, will you consider put it in others pick jobs?

Makes sense – I’ll triage this request as an addition to the manual picker in CryoSPARC and in Live, as well as in Inspect Picks.

Unfortunately this isn’t possible because the power/NCC score are metrics that evaluate how well a given template (from the template, blob, or filament pickers) match the particle image. These metrics don’t exist with manual picks, because there is no reference template used to select the manual picks.
