Focused refinement in future versions?

Hi all,

I was wondering if there are any plans to include focused refinement functionality similar to the one found in Relion in the future? If so, any idea when this feature could be available?

I am working on viruses and other highly symmetrical complexes with oftentimes large differences in
flexibility of certain domains, so the ability to carry out focused refinements of certain parts of highly
symmetrical objects would be extremely useful.

I know of and have used the Local Refinement feature in combination with Particle Subtraction, Symmetry Expansion and Masks, but the results are not particularly convincing so far.

Hi @twg - I have used local refinement in combination with symmetry expansion in cryoSPARC and in my experience so far the results are comparable to Relion. I don’t usually perform signal subtraction as mostly (with exceptions) I have found it of marginal benefit. What is the exact workflow you’ve been using (that you haven’t found effective) - perhaps there are some parameters you could tweak?
