Could not get image from topaz denoise in manual picking

Hi,

I processed my micrographs using Topaz Denoise, but when I attempt manual particle picking in CryoSPARC, I encounter an error: “Could not get image for micrograph.” The micrograph information is present, but I am unable to open them for particle picking.

After investigating the issue, I discovered that the pixel value is mismatched between the input micrographs and the denoised output.

I would greatly appreciate any guidance on resolving this issue.

For reference, I have attached a screenshot of the manual particle picking error.

Hi @Amy @rkyadav0889 have you tried the CryoSPARC Micrograph Denoiser? We generally find it produces great denoising results

I don’t think this is directly Topaz related, but a bug in the Viewer which sometimes pops up even with CryoSPARC’s own denoiser. I’ve never worked out what specific sequence (Patch Motion/Patch CTF/Denoise/Curation or different order) is guaranteed to trigger it.

@rbs_sci Next time you encounter the error, please can you check under Metadata|Log for any errors.
