Hi.
I am also dealing with small particles of around 60 A diameter. I am new for this technique. I got the 2D classes as below. Can anyone suggest if these are good or not?
These look like noise. What do your micrographs look like?
Hi Stefan,
I also caculate a small size particle recently. You make a great progress in your sample. Can you show me the key parameters you use in your case. The size of my sample is around 60 A, and the pixel size is 0.743, defocus is 1.0-2.5 um, I set the box 288 A. For 2D, I use number of iterations and batchsize (40/200), and set “Force/max over poses/shifts” to “ON”. Also I tried set “Force/max over poses/shifts” to “OFF”, it seems worse.
The feature is blurred.
Hope to get recommadation from anyone here.
Thank you
Qingpeng
Hi Jamon,
I also got the same problem with you. Have you resolve the problem finally?
Hello Olib,
I got a dataset of a small size proteins. I post my results and the parameters for my 2D classification. The pixel size is 0.743, boxsize is 320, Could you please give me some recommendations? Thank you so much.
bad picking? inspect picks and try to get 1 pick per dark feature. use slider bar, use template picking (even providing a few of the terrible templates from your first 2D job).
box too big? impossible to tell with zero feature alignment, but would suggest to try half the box size at least at start to get alignment.
