I am now processing a helical dataset (amyloid) in both cryoSPARC and relion, and have one thing I do not really understand. When I performed both manual pick and topaz auto-picking in relion5, the tubeIDs of the segments from the same fibril are the same. For example, I have tubeID 1, 2, 3 for the segments from three different fibrils in image 1. but in the image 2, the tubeIDs restarted from 1 again when I did the same picking, ie 1, 2 ,3… in this case, I have segments with the same tubeID, but they are from different fibrils from different images. As I know the tubeID should be specific for segments from the same fibril. My question is, should I reassign the tubeID in particle.star file to make the segments from the same fibril have unique tubeID, ie the IDs in image 2 should start from 4, 5, 6…? thanks for the helps.
