I’m wondering (stimulated by recent discussion on 3DEM board) whether it might be possible to add a variant of the local-FSC based local resolution tool to analyze local differences between conformational classes? Seem like a local FSC volume might be handy for comparing classes after heterogeneous refinement or ab initio reconstruction.
Only thing extra required (I think) would be the capacity to automatically align volumes prior to calculating local FSCs (and maybe the option to locally align each subvolume used in FSC calculation, in order to pick up on more subtle local differences that might be lost if not extricated from the movement of larger domains).
