Set the class number of output

Hi specialists,

My data particles are heterogenous very much. The averaging particles are different in size and have different shapes. It is hardly to seperate them from 2D classification. So I want to try “ab initio reconstruction” or “heterogenous refinement”. But I’m not sure where to set the class number for both experiments. I tried several options, but finally, it ended up with only 2 maps, which is unexpected from the 2D classification.
I am not sure how many conformations are mixed in my data. So I want to tried a initial number of 20 for classes setting in experiments. Maybe it will end up in 6~8 classes. Could you help me to set this? Thank you very much!

Jianhao

Hi @jianhao
Please see this post on ab-initio reconstruction which shows how to select a certain number of classes.
