Dear Cryosparc-Team,

I was looking for a satisfying solution to include the Falcon 4i defect file. but could not find one or the case was not further followed. I can provide you our defect file and would appreciate feedbeck on how to convert it into a format cryosparc can understand. Please let me know to whom I can send the file for inspection.

Please, I am not looking for a solution to use the gain-file in order to feed the defect file information, but a solution on how to convert our defect file format/ get to know the required format.

A simple 4 column text file is all that is needed, e.g.:

3183 3253 3 3
3189 3435 1 1
1 3406 4096 1

Format is: [x start][y start][pixels to step in x][pixels to step in y]. For more information, see the MotionCor2 manual. If you need an exhaustive list of known defects, you can check in the Falcon 4(i) Service Tool on your microscope, under the Sensor Defects tab, although the formatting looks different.

Until Thermo fixed some issues with how they output their .gain files, we needed to use the above to fix some defects on one of our sensors. With newer versions of EPU, we only need the .gain file.

Our defect file basically looks like this:

Which means, that I may a few more lines than you probably, right? :smiley:

Accordingly, a defects file which cryosparc understands, shuld look like this?:

3596 0 1 4096
1217 0 1 4096

That seems about right… :slight_smile:

You might need to flip gain/defects in Y.
